This intermediate is N tosylated and then subjected to a Mitsunobu reaction to invert the hydroxyl group, followed by condensation with 2,2 diphenylacetonitrile to build the substituted pyrrolidine core. The tosyl protecting group is removed under acidic conditions to give the corresponding pyrrolidine salt, which is then coupled with 2 (2,3 dihydrobenzofuran 5 yl)acetic acid to form an amide. Further, the amide is reduced to the free amine, converted to its hydrobromide salt, and then hydrolysed under basic conditions to form the acetamide intermediate. After crystallisation, the final reaction with hydrobromic acid results in the formation of darfenacin hydrobromide as the finished product.

Darifenacin is a selective muscarinic M3 receptor antagonist used for the treatment of overactive bladder with symptoms of urge urinary incontinence, urgency, and frequency. It appears as a white to almost white crystalline powder. The molecular formula of the compound is 
C28H30N2O2, and its molecular weight is 426.5 g/mol. The compound has a melting point of 228 to 230 degree Celsius and a boiling point of 614.3 degree Celsius at 760 mmHg. The compound is soluble in water at less than 1 mg per mL at 25 degree Celsius, and is also soluble in chloroform, DMSO, methanol, and ethanol. It is marketed under the brand names Enablex and Emselex. It may cause serious health problems, including severe constipation, urinary retention, blurred vision, dry mouth, heat prostration due to decreased sweating, gastrointestinal disturbances, and, in rare cases, severe allergic reactions and angioedema. It is contraindicated in patients with urinary retention, gastric retention, or uncontrolled narrow-angle glaucoma, and the medication should be stored at 4 degree Celsius protected from moisture.
