In the final step, the DMF solvate is reacted with triethylamine in i-PrOH, followed by treatment with sodium 2-ethylhexanoate, which leads to the formation of Fostamatinib hexahydrate as the final product.

Fostamatinib is a spleen tyrosine kinase inhibitor used in the treatment of chronic immune thrombocytopenia in adults. It appears as a white to off-white powder and is formulated as the disodium hexahydrate salt in pharmaceutical preparations. The molecular formula of the base compound is C23H26FN6O9P, and its molecular mass is 580.47 g/mol. The compound is practically insoluble in a pH 1.2 aqueous buffer, slightly soluble in water, and soluble in methanol. The melting point of the compound ranges between 214 – 217 degree Celsius, and its boiling point is around 814.2 degree Celsius. It has a density of around 1.496 g/cm3.
