Product Overview

The report provides a detailed analysis essential for establishing an Ingenol production plant. It encompasses all critical aspects necessary for Ingenol production, including the cost of Ingenol production, Ingenol plant cost, Ingenol production costs, and the overall Ingenol production plant cost. Additionally, the study covers specific expenditures associated with setting up and operating an Ingenol production plant. These encompass production processes, raw material requirements, utility requirements, infrastructure needs, machinery and technology requirements, manpower requirements, packaging requirements, transportation requirements, and more.

Ingenol is a diterpenoid derived from Euphorbia peplus sap. It is mainly utilised in pharmaceuticals as the core structure for derivatives like ingenol mebutate (Picato gel), which is FDA-approved for treating actinic keratosis, a precancerous skin condition, by activating protein kinase C to induce rapid necrosis and immune responses in abnormal cells. These topical formulations, available in 0.015% and 0.05% concentrations for face/scalp and body, respectively, offer sustained lesion clearance.

Additionally, ingenol derivatives have shown potent antileukemic, anti-HIV, anticancer, and anti-inflammatory activities in research, with improved analogues like ingenol disoxate showing enhanced stability for non-melanoma skin cancer treatments and potential ambient storage.

The ingenol market, primarily driven by ingenol mebutate (Picato Gel), is propelled by the rising prevalence of actinic keratosis due to ageing populations and increased UV exposure. Regulatory support from the FDA and EMA, including streamlined approvals for topical therapies, as well as growing healthcare expenditures and insurance coverage, enhances accessibility and demand for minimally invasive treatments such as ingenol.

Additionally, emerging trends like AI integration for diagnostics, sustainable production, and expansion into high-growth regions such as the Asia-Pacific further boost growth. Industrial Ingenol procurement for pharmaceutical use is influenced by its natural extraction from Euphorbia plants, requiring complex chromatography purification or synthetic routes from precursors like (+)-3-carene, leading to limited suppliers and high production costs.

Raw Material for Ingenol Production

According to the Ingenol production plant project report, the raw material for Ingenol production includes allyl ether.

Production Process of Ingenol

The extensive Ingenol production cost report consists of the following major industrial production process:

  1. Production via the Tanino–Kuwajima racemic total synthesis: The production process of ingenol starts from allyl ether via Claisen rearrangement to form a ketone compound, followed by NBS bromohydration to form the first intermediate. In the next step, the intermediate undergoes aldol/dehydration to get enone, followed by its intramolecular alkylation with Me3Al/LDA to give trans-decalin. The resulting compound is subjected to side-chain extension to provide dicobalt acetylene complex, followed by cyclisation to obtain allyl alcohol using methylaluminum bis(2,6-dimethyl-4-nitrophenoxide). The next step involves Birch reduction/dibromocyclopropanation/methylation for the D ring, and epoxy alcohol rearrangement to form the key intermediate compound. In the next step, AB ring functionalisation then proceeds through enone formation, dehydrations, stereoselective OsO4 dihydroxylation of diene to polyol, cyclic carbonate protections, and dehydration to yield the olefin. Finally, Corey-Kim oxidation forms α-ketol, followed by epoxide formation, Zn/NH4Cl reductive cleavage, and hydrolysis, achieving ingenol as the final compound.

Properties of Ingenol

Ingenol (C20H28O5) is a tetracyclic diterpenoid with a molecular weight of 348.44 g/mol, having a complex fused ring system including a cyclopentenone, multiple hydroxyl groups (at positions 5,5a,6 and a hydroxymethyl at C4), and four methyl substituents (1,1,7,9). It exists as a white to off-white solid, sparingly soluble in water but soluble in organic solvents like DMSO, methanol, and chloroform, consistent with its polyhydroxylated yet lipophilic polycyclic structure. It is a cyclic terpene ketone with potent biological activity derived from Euphorbia plants, prone to esterification (e.g., ingenol mebutate, C25H34O6, molecular weight 430.54 g/mol, XLogP3 2, 3 H-bond donors) and sensitive to oxidation or base due to its enone and vicinal triol moieties.
