Nafcillin is a semisynthetic, narrow-spectrum penicillin antibiotic that appears as a white to off-white crystalline powder. It has a molecular weight of 414.5 g/mol for the free base and 436.5 g/mol for the sodium salt monohydrate. It is stable under refrigerated conditions as a lyophilised powder, and it features a LogP of around -1.9, indicating moderate lipophilicity. It has a polar surface area of ~96 Ų, and collision cross-sections in mass spectrometry ranging from 180–202 Ų for various adducts. It contains a beta-lactam ring resistant to staphylococcal penicillinases, a 2-ethoxy-1-naphthamide side chain enhancing stability, and four rings in its structure. It goes through hydrolysis in acidic environments or by beta-lactamases from certain Gram-negative bacteria.
