Product Overview

The report provides a detailed analysis essential for establishing a sutimlimab production plant. It encompasses all critical aspects necessary for sutimlimab production, including the cost of sutimlimab production, sutimlimab plant cost, sutimlimab production costs, and the overall sutimlimab production plant cost. Additionally, the study covers specific expenditures associated with setting up and operating a sutimlimab production plant. These encompass production processes, raw material requirements, utility requirements, infrastructure needs, machinery and technology requirements, manpower requirements, packaging requirements, transportation requirements, and more.

Sutimlimab is a humanised monoclonal antibody that targets the classical pathway of the complement system. It inhibits haemolysis in cold agglutinin disease (CAD), a rare autoimmune haemolytic anaemia triggered by cold temperatures. It is FDA-approved for treating haemolysis in adults with CAD, reducing the need for red blood cell transfusions. It is utilised regardless of prior transfusion history, as evidenced by rapid haemoglobin increases, bilirubin normalisation, and fatigue improvement in the phase 3 trials. Its common side effects include respiratory and urinary tract infections, headache, hypertension, dizziness, fatigue, peripheral oedema (swelling in lower legs or hands), arthralgia (joint pain), cough, nausea, diarrhoea, etc.

The market for sutimlimab is driven by growth in the cold agglutinin disease treatment. The regulatory expansions covering both transfusion-dependent and independent patients contribute to its market growth. The rising rare disease awareness and orphan drug incentives boost its demand as a first-in-class complement inhibitor. The industrial sutimlimab procurement is driven by speciality pharmacy and hospital infusion centres due to IV administration and monitoring requirements. The high pricing is offset by orphan reimbursement pathways and patient assistance programs, along with limited supplier competition under patent exclusivity, which affects its market dynamics. Its global supply from biologic production hubs, and regulatory hurdles like REMS for infusion risks, alongside payer negotiations impacts its sourcing.

Raw Material for Sutimlimab Production

According to the sutimlimab production plant project report, the key raw materials used in the production of sutimlimab include Chinese hamster Ovary (CHO) cell line and solvents.

Production Process of Sutimlimab

The extensive sutimlimab production cost report consists of the following major industrial production process:

  1. From CHO cell line: The production process of sutimlimab involves engineering the CHO cell. The cells are grown in shake flasks, then in larger bioreactors using animal-free, chemically defined media at controlled pH, temperature, and oxygen levels. After culture, the broth is clarified by depth filtration and centrifugation. Its purification includes Protein A chromatography to capture the antibody, followed by cation exchange, hydrophobic interaction, and anion exchange steps to remove impurities. It goes through low pH hold for viral kill, virus filtration, and ultrafiltration and diafiltration to give pure sutimlimab.

Sutimlimab has a molecular formula of C6436H9938N1700O2013S46 (non-glycosylated form) and a molecular weight of approximately 144,800–147,000 Da. It comprises two heavy chains (every 445 amino acids, ~48,778 Da) and two light chains (every 216 amino acids, ~23,655 Da). This is linked by disulfide bonds, with N-linked glycosylation at Asn-295 and an engineered L248E mutation in the Fc region to minimise effector functions. It appears as a colourless to light yellow liquid concentrate (50 mg/mL) for IV infusion. It contains buffers like sodium phosphate, sodium chloride, polysorbate 80, and water for injection. It is stable under refrigerated storage with a terminal half-life of about 21 days due to target-mediated disposition.
