Frequently Asked Questions

The main end-use segments are oral antibiotics for mild to moderately severe infections, prophylaxis for recurrent rheumatic fever and spleen disorders, and veterinary medicine. Oral antibiotic use accounts for an estimated 80–90% of demand, prophylactic use accounts for around 8–15%, and veterinary medicine accounts for about 2–5%.
Penicillin V is produced via biosynthesis through submerged aerobic fermentation using Penicillium chrysogenum mould in large stainless steel fermenters. The process involves a liquid culture medium containing nutrient sources and the precursor phenoxyacetic acid, followed by mycelium filtration, extraction from the broth using organic solvents at a low pH, and precipitation to obtain Penicillin V. The key precursor is phenoxyacetic acid, while the fermentation medium supplies lactose, glucose, corn steep liquor, nitrogen sources, minerals, and other nutrients.
Key requirements include fermentation medium components such as lactose, glucose, and corn steep liquor, the phenoxyacetic acid precursor, and solvents such as butyl acetate, KOH, and NaOH. These inputs are estimated to account for around 55–70% of total raw material costs, with phenoxyacetic acid and fermentation media forming major cost components.
Manufacturing costs are strongly influenced by bioprocess inputs and the need to add phenoxyacetic acid, which is a major component of the production cost. The cost of fermentation media is also linked to energy costs and agricultural commodity prices for corn and sugar. Fermentation yield, oxygen and cooling requirements, solvent consumption, downstream recovery, purification, labor, and waste treatment also affect overall manufacturing costs.
Penicillin V typically appears as a white crystalline powder and is often used as the potassium salt. Pharmaceutical-grade Penicillin V potassium generally requires high assay and purity, controlled related substances, residual solvents, moisture, microbial quality, and compliance with relevant pharmacopeial specifications. It is mainly supplied as an API for oral antibiotic formulations.
Manufacturing must meet stringent pharmaceutical Good Manufacturing Practices (GMP) and regulatory requirements. Facilities require validated fermentation and purification processes, quality control testing, impurity and microbial controls, stability testing, proper documentation, and controlled handling of pharmaceutical materials. Depending on the target market, FDA or EMA requirements, DMF submissions, and applicable pharmacopeial standards may also apply.
