Frequently Asked Questions

USD 15 million to USD 50 million. This range depends on whether fermentation of the rifamycin starting material is included, the level of cGMP compliance, the number of reactors, the semisynthesis and purification scope, and whether finished-dose lines are added.
About 1 to 15 metric tons per year of rifabutin active ingredient.
The global rifabutin market size is expected to be around USD 100 million to USD 250 million and is forecast to grow at a compound annual growth rate (CAGR) of 4.5% to 5.0% over the coming years.
Mycobacterium avium complex disease accounts for 45% to 55% of demand, tuberculosis with HIV accounts for 30% to 40%, and other mycobacterial infections and combination or second-line uses constitute the remaining share.
The process involves the chemical modification of rifamycin S with an isobutyl piperidine amine building block to form the spiro imidazo piperidine ring. This is followed by purification through extraction and chromatography or crystallization, and drying to achieve pharmacopeial purity.
The rifamycin starting material and the isobutyl piperidine amine building block together represent 55% to 70% of total raw material expense.
Core process equipment, including the fermentation and recovery train, purification and chromatography systems, and crystallization, filtration, and drying units, represents 40% to 50% of the total rifabutin plant setup cost.
Manufacturing costs are mainly driven by the price of rifamycin, semisynthesis yield, reagent costs, purification requirements, product purity, and plant scale. Higher yields and efficient purification can reduce the cost per unit of API.
Capacity is concentrated in India, which is a major generic producer; Italy and other European countries, which are associated with the origin of the rifamycin class; and China, which supplies rifamycin starting materials and active ingredients.
Rifabutin is supplied as the free base and must meet pharmacopeial purity requirements under cGMP conditions. The USP monograph specifies rifabutin content of 950 to 1020 µg/mg on an anhydrous basis, along with applicable identity and quality requirements.
Manufacturing must comply with cGMP requirements and meet applicable pharmacopeial quality standards, including controls for identity, strength, purity, and impurities. For the US market, API manufacturers must comply with applicable FDA requirements, including establishment registration and drug listing where applicable. Regulatory submissions such as a DMF or equivalent active-substance documentation may also be required depending on the target market and filing route.
